I didn't know that there was a difference between road and MTB 10 speed.

I'm running Dura Ace 9 and 10 speed thumbies on two of my bikes with dura ace chains, Tiagra mechs and mtb cassettes without any trouble.

That is because you are using road shifters and mech . The new 10spd mtb groupset have a different cable pull .
 
Ok. Just tried the following set up...

Shimano 10spd Dura Ace shifter on pauls mount
Shimano 2011 XT 10 speed shadow rear mech
Shimano XT 12/36 cassette
Shimano dura ace/ XTR chain

So road shifter with mtb mech and cassette.

Worked perfectly up and down the shifts even under load.

So I can't see how there are any compatibility issues.

Just a thought on rear mechs. On my Cotic i wanted an oldskool look, so as well as thumbies I used a road mech. Its a medium cage so easily handles a 32t rear sprocket (even though the big S says 30t max). I chose the Tiagra mech because it it looks rather retro compared to a modern Shadow MTB mech, is only 20g heavier than an XTR and costs less than half that of a current XT mech. Lots of racers used 105 shortcage mechs BITD which always stood up to the rigours of mountain biking, so no reliability issues when used offroad.
So anyone looking to use these modern thumbies on a build, why not save yourselves some money and go for a lower end road mech?

Dunno where the compatabilty issues come from - Pauls and SJS may not agree?

I have SJS 9-speed Dura Ace thumbies running XTR (rapid rise rear) MT960 mech's on my 2K Explosif, XT M750 on my '97 Explosif and Pauls 7-speed Dura Ace running XT M737 on my e-stay project without any problems at all... 8)

microSHIFT thumbies certainly look the part...
 
You could use this all shimano setup:

10 speed road shifter
10 speed MTB cassette
9 speed MTB rear derailleur

Should work fine.

No idea how or why Dr S's setup works, it shouldn't!
